Twyman made one start and three substitute appearances for Gloucester in 2008/09 after graduating from the youth team, joining the Tigers from Cheltenham Town.
Jody Bevan's Fairford, who are fourth in the Hellenic Premier League, beat Abingdon Town 5-1 in the first qualifying round of the Vase, while Shortwood thrashed Tuffley Rovers 5-0.
There is £ 725 in prize money up for grabs for the winners at Cinder Lane, a place in the first round proper on October 12.
